Synthesis of a Paramagnetic Myeloperoxidase Substrate with Superoxide Dismutase Activity

ثبت نشده
چکیده

A scheme was constructed for synthesizing a magnetic resonance imaging (MRI) contrast agent with both myeloperoxidase (MPO) sensing and superoxide dismutase (SOD) therapeutic ability. This contrast agent was designed by modifying a previously published paramagnetic SOD mimetic with therapeutic ability to contain a substrate group for MPO. This allows the contrast agent to accumulate in sites of high MPO concentration, increasing MRI signal intensity. The scheme relies on economic dipicolinic acid as a starting material for the functionalized ligand. The reaction intended to be utilized to attach the MPO substrate is an environmentally-friendly “click chemistry” reaction. Both synthetic building blocks for the click chemistry reaction were synthesized and characterized and a new, final scheme was devised in order to increase yield and convenience.

Nitrite as a substrate and inhibitor of myeloperoxidase. Implications for nitration and hypochlorous acid production at sites of inflammation.

Myeloperoxidase is a heme enzyme of neutrophils that uses hydrogen peroxide to oxidize chloride to hypochlorous acid. Recently, it has been shown to catalyze nitration of tyrosine. In this study we have investigated the mechanism by which it oxidizes nitrite and promotes nitration of tyrosyl residues. Nitrite was found to be a poor substrate for myeloperoxidase but an excellent inhibitor of its...

Role of the superoxide anion in the myeloperoxidase-mediated antimicrobial system.

An antimicrobial system has been previously described in the polymorphonuclear leukocyte which consists of myeloperoxidase, H202, and a halide. In the present study, H202 is replaced by xanthine and xanthine oxidase in the isolated myeloperoxidase-mediated antimicrobial system and the contribution of the superoxide anion (02T) and hydroxyl radicals (*OH) to the antimicrobial effect investigated...

Chemiluminescence and superoxide production by myeloperoxidase-deficient leukocytes.

The role of superoxide anion- and myeloperoxidase-dependent reactions in the light emission by phagocytosing polymorphonuclear leukocytes has been investigated using leukocytes that lack myeloperoxidase, inhibitors (azide, superoxide dismutase), and model systems. Our earlier finding that oxygen consumption, glucose C-1 oxidation, and formate oxidation are greater in polymorphonuclear leukocyte...

Oxidative Status Imbalance in Patients with Metabolic Syndrome: Role of the Myeloperoxidase/Hydrogen Peroxide Axis

The present study evaluated the cardiometabolic and redox balance profiles in patients with Metabolic Syndrome compared to apparently healthy individuals, and the participation of the myeloperoxidase/hydrogen peroxide axis in systemic lipid peroxidation. Twenty-four patients with Metabolic Syndrome and eighteen controls underwent a full clinical assessment. Venous blood samples were collected f...

Superoxide-dependent oxidation of melatonin by myeloperoxidase.

Myeloperoxidase uses hydrogen peroxide to oxidize numerous substrates to hypohalous acids or reactive free radicals. Here we show that neutrophils oxidize melatonin to N(1)-acetyl-N(2)-formyl-5-methoxykynuramine (AFMK) in a reaction that is catalyzed by myeloperoxidase. Production of AFMK was highly dependent on superoxide but not hydrogen peroxide. It did not require hypochlorous acid, singlet...
